Native Instruments Massive, Diminsion pro and Firebird is a great combination for me. Those are my top favorites, with the AAS collection 'VERY' close behind.

Man. I would pretty much agree with everything that bengeorge said, except I would add Surge to the payware and I have never tried the addictive drums demo. And maybe reaktor too but only because I own it, hasn't yet been incredibly useful.

Image-Line DirectWave!
If you have to do any real sampling, you know that most "softsamplers" are a cruel joke. DirectWave is all go and no show, it's got a pretty minimal utilitarian interface. But if you need to do some actual sampling, it's a life saver. And it sounds great.
The onboard effects are very good (esp. reverb) and it's stable and CPU efficient.
