Hello,
I have a one minute recording of a siren with a roughly 10 second start, 40 second center, and 10 second end. I'd like to make the center period the sustain loop and the end of the sound a release "loop" that plays only once. Is there a way to make sample playback jump to the start of the release loop (skip the rest of the sustain) when a MIDI key is released?

No. In general, this will not work, because unless the sound is very uncomplicated, there is no way you could do that without generating a discontinuity in playback, i.e. the sound would distort/click.
If you want the release portion of the sound to only play once, btw, you can just set release to "none", and it will play non-looped after release.

You can also try layering a second group with trigger on release, then either map a copy of the sample with different start, or just set start offset in sound parameters of the release group. With appropriate AEG release on the first group, and a non-instant attack on the second you could probably get a working crossfade and have it sound appropriate.
