"Unlink from preset file" has nothing to do with used samples.
When the MUX has been loaded from a preset MUX memorizes the file it has been loaded from. Eg. the auto-naming feature will use that preset file name as the name for the MUX. The "Unlink from preset file" function clears the link to that preset file and so the auto-naming will not use the file name anymore. Wiping the link may also be relevant for safety when you want to avoid overwriting the original preset file.
