Are those "All Sound Packs" sounds and instruments already on my iPhone SampleTank (full version) but just locked? I have very limited free space left after all the music apps I've installed...

All but the ones listed under "Sound Collections" are built into the app and unlocked. The Sound Collections are downloadable so they will take additional storage.

Does this mean you first have to purchase Play/Process (now at $25) and then the HD Engine (now $50)?
If not, what if one first purchases Play/Process and later wants to get the full HD Engine? Would there be any upgrade pricing?

Also this: the App Store shows Play/Process as normally costing $25 and HD Engine $100.

Sorry I didn't see the above. You do need to purchase the former in order to purchase the latter. The in-app purchasing system should tell you so if you attempt to buy the HD Engine without having the Play/Process IAP.
