What's the best platform or method to do this?

This is obviously a job for Kontakt. It is the most popular, versatile sampler on the market, with a robust scripting engine which you can use to create custom controls or an entire GUI if you feel so inclined. I use Ableton Live, and its Sampler is a very convenient, fairly flexible tool, but it doesn't hold a candle to Kontakt. And if you ever want to distribute your library, wouldn't it make more sense to do so on the leading platform?
Incomplete list of my gear: 1/8" audio input jack.

Yeah, Kontakt can do it. I've asked a few people who've worked with Kontakt about doing something like this and got the impression it's definitely not a job for a Kontakt novice, though.

We'll see about distribution; if Cowsynth ends up as popular as our bass guitars, then it might make sense to do future libraries in a popular format. I get the feeling this is something I'm just doing for myself, though, and everyone else just wants double bass samples to use as double bass samples, not as a 17-oscillator soup.

How about UVI Workstation? That's another sampler I haven't tried but I did grab it while had the mellotron library free a couple of weeks ago. I'm just catching up on installing last month's freebies...

I'll start on the double bass samples today, too. Remove anything below the fundamental, even them out, tune them, and make infinite loop versions to use as oscillators. That's the boring bulk of the work. Sticking them in a sampler and trying a hundred different ways of detuning them is the interesting part.
