i think at first subtle differences can be very hard to discern, but if you've lived with a room and speaker setup for a long while, or have been ear training yourself for a number of years, then you do begin to notice more.

if you want to go about it scientifically, then download some double blind audio a/b comparison software, and see if you can REALLY tell the difference or not. i recommend winABX, available here:
