from the manual:

Input / Sampling

To record you must load DW into a mixer channel FX slot. DW only records sound from the mixer track and FX slot where it is loaded. This means any FX in slots preceding DW will be heard (and recorded) while those after it will be heard (but not recorded).

* Monitor - Allows you to hear the mixer track that DW is loaded into. Unless this is selected you will not hear anything on the track.
* 440 Hz - This is a 440 Hz test-tone (Note A4) to aid in tuning of melodic samples.
* Record - When selected the mixer track (and FX slot position) DW is loaded into will be recorded into Direct Wave. The recording will start when audio input is received.

I don't think DW is available as a vst effect in Cubase (last I checked). But it does work if you can find a dx wrapper for it. A free one to try is Acon. I'll test it later to see how that works.. I've used the cakewalk dx vst adapter in the past.

[Edit] Just tried Acon Effect Chainer http://www.acondigital.com/us_Downloads.html but it seems to be buggy in Cubase. Crashes. But my Cubase is old so you might have better luck with it.

Another possible approach is to use Plogue Bidule. There are several ways to use Bidule. You can use Bidule as a standalone application, and simply route the audio you want to sample to a DirectWave instance's inputs, within Bidule.

If you want to sample audio into DirectWave within Cubase, you can do that too using Bidule. You can load a Bidule as a VST effect on a Cubase track's inserts. Then, route the audio to/from a DirectWave instance within Bidule.

If you aren't already using Bidule, I urge you to try it. I can't imagine life without it now, and I am a big Cubase user. Bidule is literally a "Swiss Army knife" for VST/VSTi. You can check out Bidule here:
