I usually use Vienna, but you need the creative soundcard for that. Do you have that card?

If not there is a program called Awave studio (which I use for this, and it converts 200 formats into each other nicely too), but it is not free. You might also look at synthfont which has a "VIENA" editor built after Vienna for the creative cards. And there are more options, but I forgot them.
Some samplers do nice jobs with your sample files, for example I use the Orion Platinum sampler (just if you got Orion) and if the samples are named like c3 c#3 f#4 or so it is dead easy to make an instrument out of them, with layers and all. Same is possible with the sampler built into EnergyXT1 (even though it was not specially built for that purpose) and with many others.
But like MrM mentioned, if you got a creative card somewhere, Vienna is just nice, and free.
