You can play it "normally" of course, but one of the most common techniques to use when glide is on, is to hold down one key, and then depress a new one while holding the first (of course, this should be a monophonic patch). To some degree though, this can be a cliched technique - it sounds very "70's synth" - so use it sparingly.
