I wouldn't mention Reaktor, ACE, KarmaFX, or any modular synth. They have a very traditional interface and traditional way of thinking. That's why they don't belong on this list.
Edit: Of course you can make a Reaktor ensemble that is unique... whatever!

You are getting very defensive lately. Ace is not unique because it's a modular synth. And I definitely agree with Synplant, Alchemy and Gladiator.
I would also say Omnisphere is pretty unique. Not in it's synthesis methods, which are basically just subtractive, but in the design and use. Little things like dragging envelope rhythms from Stylus RMX loops etc. makeit a little different.
Reaktor is full of them - lazerbass, metaphysical function, gaugear, the list goes on...
