I would love them to do a vocal orientated instrument - Vocalsphere (or VoxSphere). There are some nice vocal samples already in Omnisphere (and I did a bank based on many of them - Voxworks) but would love to see them develop a fully fledged instrument specialising in all things vocal (spoken and sung). I think it's really an untapped area.

Actually I was thinking the same thing but couldn't think of a clever tag. Voicesphere? Throatsphere? Ambisphere... Anyways - Thinking along the lines of a "band": Bass, GUITAR, Drums, Voice.
I'd venture to say Voice was covered in Omnisphere with the inclusion of the Spectrasonics' Voice sample libraries.
So... Guitar, maybe?
I still place all my chips on Stylus RMX as a STEAM instrument.
And there will be cake.
P.S. You do realize we're giving them a free brainstorm session here, right?

Sorry, the 'Minisphere' idea was totally mine and fully copyrighted by me in the court of ME. That's why I'm expecting my free copy(and tons of praise) when they see the light and release this.....

Folks might be interested, financially, in a series of min-Omnispheres. Stratosphere, Ionosphere, Beatosphere, complete with each patch made up of Ozone Layers®. All libraries use the OmniPlayer.
...oh wait... Alchemy already has that concept down pat.
As far as all the kids who say, "I can't afford Omnisphere, it's too expensive..." <insert additional whiney whiney here>...
All I have to say is, "Damn Kids! Get off my Damn Lawn!"
When I was a kid, I had to walk uphill both ways in the snow, carrying my Roland Alpha Juno-1, which is a non-velocity sensitive, mono-tambral, 6-voice (not infinite voice) digitally-controlled analog synth that was heavy, noisy, and didn't have USB. With a single-line 10? 12? or 16? digit readout, a menu-driven synth with chicklet buttons. And this thing cost me $600+ in 1986. What you get today for $499 still amazes me by comparison.
For that matter Omnisphere still blows away most other things in the water, even things that are produced *today*. I am baffled why people still buy Roland Jupiter-80 and Yamaha Motif whatevers. All you need is a decent controller, and Omnisphere. If they added importing your-own-samples to Omni, then I venture to say it would best NI Komplete at that point (overlooking Guitar Rig though ... and Traktor? that's not an instrument, c'mon now).
