I don't have Sylenth but the detune knob usually let's you detune the oscillators against each other while the stereo spread knob spreads the oscillators across the stereo field.
Detune can often giva you a "phatter" and less static sound because it detunes one osc down a few cents and the other a few cents up which causes beating (The phase realtionship between the oscs changes over time).
Here is an example I quickly made using Zebra, it should work very similar in Sylenth:

I used 4 oscs in this example. First they aren't detuned at all, then I start to increase the detuning and later the stereo spread.
Everything goes back to zero at the end again.
