Adam Szabo's JP6K has been a big one for me. Whenever I need a stock trance sound I can just open it up, tweak a preset and I'm set. It's like a 50 dollar Nexus with more options.

Tough choice. In the last year I own or owned the following as "major" synths:
XILS3
PolyKBii
Synthix
Diva
Aalto
Arturia SEM
Korg AE
I like them all, but if I had to choose one, I'd go with PolyKbii. Aalto is a secret weapon that isn't always used, but it's magic in the right spot. Diva sounds fantastic, but for whatever reason is mostly uninspiring. I do love them all though in their own way.

"Best" synth - man that's tough!
I think my favorite right now is ACE - it's the one I'd most likely purchase if it were hardware.
Reaktor's been mentioned, but shout-out to two specific ensembles: Photone and Carbon 2. Both are ice-cold digital and probably not real useful for many genres, but they are
frosty-ambient-heaven...
