Dubbing Over an Imported Audio File

This should be simple: add another track and arm it for Record. The specifics are different for each DAW and should be stated in the "Getting Started" section of the manual. What DAW are you using?

Import your audio onto your first track ((probably by default 'Track 1')
On your second track, on the leftmost side of the track, and just below the name of the track (probably by default 'Track 2') click, and then select 'Input 1+2' or whatever corresponds to the inputs on your soundcard. This sets the track up to 'listen' to that input. You should see the input level meters respond too.
Again on the leftmost side of the track, click on the red 'R' that will now be visible. This sets the track up to record from the selected input.
Locate the transport controls on the bottom right-hand side. Click on the red dot to start recording.
Click on the green square on the transport controls to stop recording.
