If I have 20mb of samples and then map those samples to a sampler and the sampler file says it is 20mb, do I now have 40mb of disk space taken up, or is the sampler file just referencing the 20mb of samples?

It depends on the sampler. What are you using?

DS404, cyberfree and the native Tracktion sampler.
The sampler files are always as big as the original wave folder.

When you create a smaple bank in most cases it will copy the samples (ie double the disk space). There are only a few examples where this doesn't happen. Muzys has the option to refer to external samples, and the new sfz format can be used that way too.

DS404 not only copies the samples into a bank, but keeps a copy of the samples in each sequence you save too, so make sure you only load what you need for a song. I think Tracktion does that too.
