Setup Host to Play MIDI from File and Sing in Real Time

I need to setup some host (Sequencer or DAW) to play MIDI notes from a file through some VSTi plugin, hear the melody in a headphones, sing in a microphone in the same time along with the played notes, process the singing voice via some audio (VST, etc) plugin in a near real time, and hear both the notes and my processed voice in a headphones.

Is there some guide how to setup some host to this task?

There's no canned answer for the whole kabudle. For individual broken down tasks: sure. Pick any daw (what's wrong with the one you use for some years now) and this can be done. Take time to read the manual and get comfortable with how it works. Try things out.
