I'm planning to sample my Roland TR6S and curious what KVR thinks is the best software for that. I've done a bit before using Cubase and Bitwig but for a number of reasons, this seems inefficient.

I found Reaper really useful when I did a bunch of stuff a couple of years ago. Depending on what you want to automate, you may need to get your hands dirty. David Hilowitz has a video of his working method which may help if Reaper's up your street

Check out Steinberg Wavelab, it has a batch processor as well as a scripting language to automate tasks.

In Reaper you can use a script to generate a bunch of regions along with MIDI notes, then record and render each region to a separate file. I’ve got a lua script for that at https://github.com/jonastallhage/sampling-tools
There’s also some code for finding loop points. Not sure if it’s all that user friendly though, and it’s pretty much brute force so it takes a lot of time (usually gives good results though).

Record all hits on to a single audio file (make sure there is enough space between the hits for silence)
Open in Audacity, select Analyze\Label Sounds and set Threshold level low enough so it detects the full decay (try -50)
File\Export Audio\Export to computer\Split files based on: Labels
Bonus:
Use ConvertWithMoss to convert from Wav to your preferred sample format

Don't overthink it. Any 'ole audio editor will do.

Yes but it took me about 60 seconds to make this Kontakt file with Audacity and ConvertWithMoss:
