If you want to play a sample, insert Factory -> Instruments -> Devices -> MuSampla in the MUX and connect its event in and audio out. Then open its editor and
drag-drop the sample file onto the empty sample display
or
click the sample display -> Browse -> and browse towards the sample file.
Hope this helps.

Ok, since English is not my 1st language and a picture is worth a thousand words I've made this simple video to explain 2 ways you can achieve what you want. Like I've said sorry for my bad English (I NEVER speak it, only write it) but at least if you don't understand what I'm saying you'll have a visual representation.
Also I don't have Cubase nor Maschine but I know Cubase support midi effects (or midi inserts) so you can use Cubase and Groove Agent 1.
Here's the vid:
Hope this will be useful to you and give you some new ideas too.

Thank you guys for your help. Honestly. I'll keep you guys updated,

The way Cubase and Reaper handle virtual instruments are completely different. I THINK that in Cubase (I have some experience in Sonar and I think it's the same in Cubase) you must load your vsti in a virtual instrument rack and then create a MIDI track and output the data to the vsti, is that right?
In Reaper a track can have both MIDI and audio clip, there's no distinction so you can put midi effects, vstis and vsts directly on the track.
The way I did it in the video, MUX is loaded before the VSTi (Sylenth in this case) so MUX is acting like a midi effect. The advantage of this is you can change the instrument easily while keeping MUX there, on the channel.
I know in Cubase there's a place in the track channel called "midi effects" or "midi inserts". That's where you must place MUX, if you want it to act like a midi effect (like in the video). If you want to use MUX like an instrument, read on.
If you want MUX to act like an instrument and always use the same setup (always use MUX with Groove Agent One), you can put the instrument directly in MUX, but now the difference is the output must be audio instead of event, like this:

This way you can put MUX directly in the instrument rack of Cubase, create your MIDI track, send the notes directly to MUX and you'll be ready to go!
Hope this help, if you have more questions please let me know. If this is not clear I can do more videos.
Bye!

Unfortuantly it is not possible to do it in cubase. It may be because it is VST3. Anyways, I figured it out how to do it in maschine, by using the mux sampler and going from there. Thanks you guys for all your help.
