All the things I suggest are available individually on different programs. Many programs as suggested (Sonar has a "remove silence" function) to chop it up. Many programs, including VSampler can detect pitch and map it to a key. To detect velocity should be easy as almost all programs can detect amplitudes. The only one that might be asking a bit too much is to do it al in real time.
To do this with a single sample mapped across the key range would be easy, but defeats the object. I want to create a mutlilayered soundbank / library from my own guitars that would be most realistic and possible make it available for others. Such a sampler should work just as good for flutes or other real instruments.
