how can I use vst effects in realtime, i. e. got my guitar connected to my soundcard - and now?!

I dont think there is a wider question you could have asked apart from how long is a bit of string!!!
OK, so you will need some HOST software to host the VST / VSTi you want to use... TobyBear has a ShareWare one called MiniHost that is a lot of fun - check it out using the quick links on the top right corner of KvR...
With guitar you should be aware of latency - the delay between playing your guitar / instrument and hearing sound. The better your soundcard - the lower the latency you can have... but beware low latency can be processor intensive.
Sorry if you know all this and already have a host and some vst / vsti's - as I said its a pretty broad question

I'm fine with the host/vst/vsti-side, I just really can't figure how to connect in realtime with fruityloops.
(ok, started as an effect topic, is moving slightly to the hosts
).
(and yes, I'm having a lot of fun here already, I read more than I write (what is a good thing sometimes))

Ah fruity - OK!
I had the same problem myself awhile back... Umm... OK...
Set Fruity to SONG mode.
Pick any Mixer Channel / say FX channel 4 in the mixer.
Set your output to master.
Set your input to your soundcards in -> mine is Line In1 or Mic In1 on my LapTop SB card.
Click the REC icon on the bottom right of Fruitys mixer (this lets you specify a filemane and location for your recording).
In slot 1 of your channel - insert some FX.
Use the knob to get a wet/dry balance.
Theoretically - you should be able to now play and hear your fx in realtime, even without actually recording anything!?!?!
