Markers in Acidized files...some advice please

Could someone explain the process of setting markers in files? If I wanted to place markers in a 4/4 percussion wav file, do I just add those markers at the "2" and the "3", since the beginning and end of the file are already defined? (Assuming a one-bar file).

Also, from what I've read on this forum, this seems to really make a difference in the file's "stretchability" without artifacts...in other words, you can go even further up or down in tempo, without artifacts, than with a "simple" acidized file. Thanks in advance for sharing your time/knowledge in this.
