The setup:
Cubase SX 3.1 connected to Gigastudio 3.12 with Rewire
Windows XP SP2
Processor: AMD Athlon 64 X2 3800+ (so it's a dual-core)
I can't use GSIF(2) as my soundcard supports only ASIO.
The problem:
While playing back or bouncing to disk (audio mixdown) Gigastudio tracks in Cubase
some notes seem to disappear (not cut off, the notes don't get played at all!).
I've had the problem with many earlier Gigastudio 3.x versions
and also Cubase SX 3.0, but I'm now using the latest versions...
I do audio mixdowns faster than real-time, but as the issue is there
also with normal playback in Cubase this can't be the reason for the problem.
I usually use Gigastudio for piano sounds so it gets really annoying
when you suddenly lose pedal or some notes of the melody (for some reason it
seems to be the pedal that gets cut out very often).
I have no problems playing stuff live (from a midi keyboard) with Gigastudio samples, so the
note drop is only related to playback.
Also, this has nothing to do with insufficient polyphony as there can be only 3-4 notes
active and I still get these note drops.
EDIT: To be more clear with the "symptoms": it seems like some MIDI data just vanishes
on its way from Cubase to Gigastudio as if a note reaches GS and gets played, there seem
to be no problems with the sound or anything like that.

I was told there was "issues" with GS3 and dual core cpus. What they are, I have no idea. You might want to try setting GS3 to only run on one core. You can do this by openning up Task Manager, right clicking on the GS3 process (not sure what it will be listed as), choose Set Affinty and then unclick one of the cpus.
This is completely a shoot in the dark and I have no idea if it will work. I have SX3 and GS3 Ensemble but I haven't installed GS3 yet just sitting there getting dusty.

Well, I don't think this is the case now, because, as I said, playing live works perfectly.
So if there was a problem related to multiprocessing in GS3 I think it would affect all playing.
(And the changelogs of 3.x versions indicate that a number of problems related to multiprocessing/hyperthreading have been fixed)
Anyway that could be worth a try, but all other comments are very welcome...
I'd finally like to have a working Cubase3<->GS3 connection.
