to make those really fast drum rolls and stutters as well as to stutter vocal samples is it best to use some sort of effect or sampler or is the best way to do it to cut up a sample manually in 32 or 64ths

Yeah, you can chop them up manually but a plugin like Lucifer http://devine-machine.com/ is way easier to use and much more expressive.
