I'm a Spire user for a not very long time, so it's possible I do something wrong. The sequencer is playing long sustained notes, ENV3 is modulating Cutoff1. Now I turn the amount knob and it sounds like that the filter envelope gets retriggered (without new Note-on message). Is this how it should work? I do this on many synths for years, but I never had this special behaviour. I need advice.

Check what is modulating the filter then, there must be some other source like LFOs or ENVs or the Arp is active?

It's the Init patch, there is no additional modulation. But to be safe, I set everything to zero, also the Amp levels of the LFOs and I cleared the Mod Matrix completely.

During making this screen recording, I noticed the repeating is related to the buffer size of the audio driver. Small size (512 samples) fast repeating, big size (2048 samples) I get this result....

Unfortunately, the knobs related to the modulation matrix are not intended to be automated. Because recalculation goes through the buffer, not sample-by-sample, this saved the load on the processor.

Thank you for clarification, that's surprising. But, at least for Ableton Live I already have a workaround. There is a Midi FX device called Velocity, it's not exactly the same as modulating the (Filter) Envelope Amount knob, but at least no repeatings. To work properly, all four knobs of ENV3 must be turned to the right.
