With lots of transpiration and a tiny bit of imspiration: trying out different things.

If there's just one monophonic melody, then the sky is basically the limit.
If your existing vocal track already contains a second line, then there's already more harmonic context you have to take into account. Could make it easier, or could make it more difficult. It depends. Take the existing notes and think of chords that already contain those notes. You only need to add a third note to construct a basic chord.

Now listen to Bobby Brown's "Two Can Play That Game". It's a fine example of what can go wrong if you whack new chords to an existing melody. If you don't hear the dissonant notes, then no worry. It still can peak the charts! If you do hear where it's wrong, then you know what to avoid ;-)

Unless it's Rap it's unlikely that all the vocal is on one specific note.
It's more likely that Melodyne is set for percussive material and not melodic. I forget which menu it is in, but it should be easy to find

doh !!

Some possible starting chords: C, F, Am, Em.
More possibilities than you can handle ;-)

excellent !!, ill maybe just try some basic stuff to see what fits,

not very easy playing around vocals, much prefer to create a progression and have the vocal added later

If you have troubles finding chords, then you can try the following.

First analyse which notes are used in a small section of the melody, say one or half a bar. Underline which notes feel more dominant than the others. Those are candidates to construct triads from. Can be a bit of a puzzle...
