If there's just one monophonic melody, then the sky is basically the limit.
If your existing vocal track already contains a second line, then there's already more harmonic context you have to take into account. Could make it easier, or could make it more difficult. It depends. Take the existing notes and think of chords that already contain those notes. You only need to add a third note to construct a basic chord.
Now listen to Bobby Brown's "Two Can Play That Game". It's a fine example of what can go wrong if you whack new chords to an existing melody. If you don't hear the dissonant notes, then no worry. It still can peak the charts! If you do hear where it's wrong, then you know what to avoid
