in the FX Grid, i place a delay module in lane 2 and turn up the lane 2 level, i play some notes with an oscillator into the "main" lane......so i have th audio signal going through the main lane......
my question is: what is the difference between rightclicking the delay module and selecting input 1 as the input VS just turning the knob for send 1 up
they both seem to have the same effect.....which is to send the lane 1 audio into lane 2 of the FX grid

In your example they are exactly the same. By default all 4 lanes of the main grid above all send their stereo summed output into the main (left-most) lane of the FX grid. That's what the circled boxes under the main grid elements indicate so:

If you change one of those to "bus1" or "bus2" they will be send instead directly to bus1 (the middle FX lane) or bus2 (the right-most FX lane).
Now, let's take a look at a more complicated patch. In the factory soundset open up 0 Tenth Anniversary -> HS Bamboo Work.
I've indicated with arrows the complex routing this patch utilizes:

Here both lanes of the main grid are routed into "bus 1", the middle FX lane. From here Howard applies a compressor and reverb to the signal. Next he splits the signal into two separate equalizers and mixers in the left and center lanes. He wouldn't be able to do this without the ability to select "Input 2" for EQ1 in the left-hand lane.
And that's what the menu option you asked about is for: more complex signal chains when routing in the FX area.
