MTuner audio input?

How do I select the audio input for MTuner to tune instrument (external synth)?

What do you mean? You just insert it on your track in your plugin host.

When inserting MTuner onto an audio track in Logic or Live with track's external inputs assigned to external synth/guitar there is no meter activity. Apparently MTuner only works with plugin synths and audio file playback.

That's something you need to figure out in your host, not the plugin. In Cubase for example one just clicks onto the "monitoring" icon.
