I want to sound the vocal sample like a lead synth, when I use MuSampla it just doesn't work I want it sounds like monophonic

Just use pitch bend on your keyboard; it'll be more humanized.
On the MuSampla, you can set the P.B. (pitch bend) to 7 (half-steps) so you won't go past G; you can then record your playing on an automation track with an Event Recorder, and tweak further.

From your description I guess you want something like the vocal leads that are used in EDM or so...
I made a quick patch , with sample player (I didn't had a vocal sample, so I used a percusion sound) inside a polysynth that is in mono, and with legato glide, also I added 2 marks in the sample player to keep the sample looping.
I hope that this is what you are looking for, and gives you some Ideas

Hmmm, I'm not sure if it would be needed, why do you want to use musampla?
You could build the equivalent using the sample player inside polysynth, and adding some envelopes, LFOs & a filter.
