Are the parameters exposed to the host? Reason 9.5 could not detect them. I haven't tried to automate Musampla in another host as I suspect the parameters for the ADSR module for instance, are just not seen.
How does it work? Are any parameters exposed to host? Is that where the 32 macro's of the MUX come in?

Indeed you can map any parameter to one of the 32 MUX meta-parameters (the top level), and those 32 meta-parameters are seen by the host.

Technically MuSampla will always be in a MUX, cause the top level is a MUX and so anything contained within it is inside a MUX. MuSampla does not need to be in the top level MUX, it can also be in a nexted 'child' MUX. Whatever the hierarchic level you can drag-drop MuSampla parameters on the top MUX meta-parameters.
