Can MDrummer plugin be unsynced from the host?

I want to experiment a bit with running different instances at different tempos. Is there anyway to specify the tempo to use for a specific instance instead of defaulting to the host?

Well, to be honest I'd rather say no to this. It is already really hard to make it work under the variety of buggy hosts, this would make it even harder and would be useless in most cases.
